Are you talking about work hardening? Hmmmmm. Shouldn't be an issue if the wheel is properly built.
Yes and no--both work hardening and residual stress as a result of earlier yielding. Fortunately, the stainless steel used most commonly in spokes doesn't harden as fast as a PH alloy or something, but it happens when you adjust the outside J-bends. (As XXXer mentioned upthread.)
